Calponin is a calmodulin, F-actin and tropomyosin binding protein, which is thought to be involved in the regulation of smooth muscle contraction. Calponin expression is restricted to smooth muscle cells and has been shown to be a marker of the differentiated (contractile) phenotype of developing smooth muscle. Multiple isoelectric variants of calponin have been identified, however only two molecular weight isoforms exist, a 34 kDa form and a 29 kDa form. Expression of the 29 kDa form, I-calponin, is primarily restricted to muscle of the urogenital tract, whereas the higher molecular weight variant has been demonstrated in vascular and visceral smooth muscle. In Western blotting, this MAb reacts with only the 34 kDa form of calponin in extracts of human aortic medial smooth muscle and is unreactive with fibroblast extracts of cultivated human foreskin. This MAb reacts with the rod domain of human cytokeratin-19 (CK19), a polypeptide of 40 kDa. CK19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. This antibody recognizes a protein of ~140 kDa, identified as N-Cadherin (NCAD), also known as CD325. NCAD is a member of the Cadherin superfamily, and consists of five extracellular repeats, a transmembrane domain and a cytoplasmic domain. CD325 deficient mice die at day 10 of gestation and embryos display major heart defects and malformed neural tubes and somites. Consistent with this, CD325 has been implicated in several aspects of cardiac development including the precardiac mesoderm, establishment of left-right symmetry and cardiac looping morphogenesis. Furthermore, CD325 is normally involved in inducing cell cycle arrest and its expression is frequently deregulated in cancer cells. Thyroid-stimulating hormone (TSH, also known as thyrotropin) is a glycoprotein involved in the control of thyroid structure and metabolism, which stimulates the release of the thyroid hormones. TSH is regulated by thyroid hormone (T3) and various retinoid compounds. TSH binds to the thyroid-stimulating hormone receptor (TSHR), which is cleaved into two subunits, A and B, and plays a major role in regulating thyroid function. The third cytoplasmic loop of TSHR has been identified as critical for its role in regulating inositol phosphate and cAMP formation. GP2 (glycoprotein 2), also known as ZAP75, is a 537 amino acid secreted protein. It is an integral membrane protein that is secreted from intracellular zymogen granules and associates with the plasma membrane via glycosylphosphatidylinositol (GPI) linkage. GP2 is cleaved and then released into the pancreatic duct along with exocrine secretions. GP2 binds pathogens such as enterobacteria, thereby playing an important role in the innate immune response. The C-terminus of this protein is related to the C-terminus of the protein encoded by the neighboring gene, uromodulin (UMOD). GP2 is also expressed on the apical plasma membrane of specialized microfold (M) cells among enterocytes and serves as a transcytotic receptor for mucosal antigens.
